How the screen was done

 

This page gives you  short overview how the LIPIDROPHE screen was performed......

In short, we treated Drosophila S2 cells with interfereing RNAi's to knock down expression of a single gene at a time. After 3 days, we induced the formation of lipid droplets by adding oleate containing medium (see also movie). After 13 h cells were fixed, stained for lipid droplets with BODIPY and for nuclei with DAPI, imaged in a high throughput microscope and scored visually and with an autmated image analysis algorithm. Potential hits were treated with a second set of siRNA's and processed the same way.....

Hits that were confirmed with an independent set of siRNA's were sorted into 5 phenotypic classes as indicated below. Images form the screen sorted according to the class they belong to can be found at the "Galleries of hits" page.
